Evolutionary Design of EEG Data Classifier

Tato diplomová práce se zaměřuje na vývoj efektivního klasifikátoru pro klasifikaci kandidátů na základě extrahovaných vlastností z elektroencefalografického (EEG) signálu. K dosažení tohoto cíle byl použit genetický algoritmus pro výběr příznaků a optimalizaci klasifikátorů na základě pěti kritérií: minimalizace počtu příznaků, minimalizace doby inference a maximalizace klasifikační senzitivity, specificity a přesnosti. Pro extrakci příznaků s cílem klasifikovat kandidáty jako trpící MDD, nebo jako zdravé, byla použita EEG data s otevřenýma očima 31 kandidátů trpících depresivní poruchou (MDD) a 28 zdravých kandidátů. Byly otestovány dva algoritmy, NSGA-II a NSGA-III. Navržený algoritmus pracoval se třemi kritérii, ale byly přidány dvě další kritéria - senzitivita a specificita. NSGA-III byl v tomto případě účinnější a byl použit v následujících experimentech. Byla zavedena omezení pro zlepšení parametrů a byly vyzkoušeny různé hodnoty pro pravděpodobnost mutace a křížení. Vygenerované klasifikátory dosáhly průměrné přesnosti 91.36 \%, senzitivity 91.82 \% a specificity 90.84 \%. V závěrečných experimentech byly nejčastěji používány kanály F3 a C3 a nejčastěji využívaným vlnovým pásmem byla gama frekvence. Výsledkem této práce jsou efektivní klasifikátory, které byly získány pomocí navrženého algoritmu, jenž využívá genetický algoritmus pro optimalizaci parametrů.
The impact of an emotional state on respiratory functions and configuration of the thoracic cage in patients with major depressive disorder, options of physical therapy interventions

This thesis looks at the influence of emotional state on chest development and configuration, lung functions and presents possible physiotherapeutic interventions in patients with depressive syndrome. In theoretical part, the thesis focuses on the issue of depressive syndrome, outlining the topic of psychosomatics and the relationships between psychological state and physical symptoms. The theoretical part also includes a presentation of the possibilities of physiotherapeutic methods, which can be used to improve respiratory and postural function. The main aim of this work was to determine the psychological state of patients both before and after individual therapies and to see if there would be improvement in the condition as well as improvement in the parameters of respiratory and postural activity due to performed physiotherapeutic methods. Within the secondary objective, potential correlations between these parameters and psychological state were described and the effect of physiotherapy on improvement of all parameters was shown. The m6A pathway at the molecular level and its role in neurological diseases

N6-methyladenosine is the most abundant modification in eukaryotic messenger RNA. This modification is reversible, thanks to a complex of methyltransferases and demethylases. The biological effects of m6 A are mediated through reader proteins. This complex mechanism of proteins contributes to many molecular processes such splicing, translation and transport. It also plays a role in many serious neurological diseases, such as Alzeheimer's disease, Parkinson's disease, major depressive disorder and attention deficit hyperactivity disorder. The purpose of this thesis is to describe the m6 A pathway, its regulation at the molecular level and to put it into context with neurological diseases of today. Circadian sleep regulation and sleep deprivation

The circadian system is designed to generate circadian rhythms and serve as the human inner clock. This is achieved through the molecular mechanism of autonomous transcriptional-translational feedback loops, in which so-called clock genes are involved. Circadian rhythms regulate the timing of physiological and behavioral processes, including sleep. Sleep is important for the proper functioning of the human organism. As a result of desynchronization of circadian rhythm, disturbances of sleep arise which threaten the mental and physical state of man. One of the examples is sleep deprivation which is deffined as a lack of necessary amount of sleep. Besides its negative effect on human health, there had been also reported positive effect in the treatment of symptoms in patients with unipolar depression. Other studies suggest that deprivation causes a reset of the circadian system, correcting the abnormal functioning of the internal clock. These effects have only a short duration, but it appears that they could be stabilized by combining sleep deprivation therapy with other therapeutic approaches. However, it is crucial to understand the exact mechanism that causes the positive effect in sleep deprivation therapy.
Antidepressant effect of ketamine

Ketamine is used as an anaesthetic mainly in veterinary medicine and paediatrics. Thirteen years ago, interest in ketamine as an antidepressant emerged. Ketamine as an antidepressant works rapidly and is also effective in the case of patients suffering from treatment resistant depression.The main disadvantage of ketamine as an antidepressant is its psychotomimetic effect. Ketamine is an antagonist of the NMDA receptor, which has an important influence in brain activity. Through antagonism of this receptor, a large amount of processes can be affected, for example levels of glutamate, excitotoxicity, or density of other receptors. In addition, ketamine probably also affects the D2 receptors, but its role in this is uncertain. Ketamine influences synaptic plasticity and its effect can be modulated by repeated application, co-application with other drugs and other methods. Also gender seems to play role in the effect of ketamine. The aim of this work is to summarise possible mechanisms of effect of ketamine, its effect on synaptic plasticity and also possibilities of modulation of its effect. This thesis mentions the most important studies concerning ketamine as an antidepressant and possible future directions of research.
